Ontwikkeling van een nieuw medicijn bij de ziekte van Graves

Eindelijk weer eens een nieuw onderzoek naar een mogelijk nieuw geneesmiddel bij de behandeling van de ziekte van Graves. Te hopen is dat dit de therapie is die de autoimmune oorsprong aanpakt van de ziekte. Iets wat welkom is.

Het medicijn (ATX-GD-59) dat onderzocht wordt is een mengsel van peptiden. Deze peptiden komen van nature voor in eiwitten in het lichaam. De bedoeling is dat dit nieuwe medicijn de immuunrespons van het lichaam verandert. Daarmee wordt de productie gestopt van de antistoffen die de schildklierstoornis veroorzaken.

Deze eerste studie bij mensen test of deze peptide therapie veilig is en goed verdragen wordt. Het medicijn is ook ontworpen om te kijken of de immuunrespons van het lichaam kan verbeteren en of het kan voorkomen dat een schildklierstoornis zich ontwikkelt.

“This is the first new therapy for Graves’ disease to be tested in over 50 years and is a significant step forward in the development of the drug,” Simon Pearce, MD, professor of endocrinology at Newcastle University, said in the release. “We are very pleased to be leading the first clinical trial of ATX-GD-59, which has significant potential to treat the underlying cause of this condition for the first time.”

DAVIAD - The development of a novel therapeutic vaccine for the selective treatment of the autoimmune disorder, Graves’ disease

This aim of the DAVIAD proposal is to develop a therapeutic vaccine for Graves’ disease (GD) that has a better safety and efficacy profile than current treatments. There is a high level of unmet need in the treatment of Graves’ disease especially in patients with Graves orbitopathy and children and adolescents (who generally suffer from the most chronic form of the disease). Autoimmune diseases arise as a consequence of a hypersensitive response to self antigens.

This proposal focuses on the development of an antigen specific vaccine therapy to reinstate immune tolerance to Graves’ disease. The selected peptides behave as apitopes (antigen processing independent epitopes) and bind to MHC class II molecules in the correct confirmation to induce tolerance. In the first instance DAVIAD will further characterise antigen specific peptides (based on the autoantigen associated with the disease) already selected using a validated discovery platform.

DAVIAD will generate translational data to support a clinical trial application, conduct disease model studies and prepare the clinical trial application. DAVIAD will design and conduct a phase I/IIa trial in GD patients to show that efficacy and safety compare favourably with current treatments and those in development. The consortium has the expertise required to effectively and efficiently take the proposed vaccine through clinical development and eventual commercialisation. A disease modifying vaccine for GD will address a serious unmet need and potentially offer a therapy that can change the way this disease is treated, offering better quality of life for sufferers and reducing burden on health services.

Apitope Announces Publication of Graves’ Disease Data in International Peer-Reviewed Journal, Endocrinology

HASSELT, Belgium, and CHEPSTOW, UK, 14 August 2018 – Apitope, a clinical stage biotech company developing potential first-in-class antigen-specific immunotherapies targeting autoimmune diseases, today announces publication of its pre-clinical Graves’ disease data in Endocrinology, one of the leading peer-reviewed endocrinology medical journals.

The publication entitled: Immunotherapy with apitopes® blocks the immune response to thyroid stimulating hormone receptor in HLA-DR transgenic mice , reports the results from a series of studies to identify peptides that can be used to treat the abnormal immune response to TSHR (thyroid stimulating hormone receptor) in Graves’ disease patients and can be found here.

The study showed that a mixture of two immunodominant apitopes® was sufficient to suppress both the abnormal T cell and antibody response to TSHR in HLA-DR transgenic mice. Tolerance induction was not disrupted by current drug treatments for Graves’ disease. The paper concluded antigen-specific immunotherapy with apitopes® from TSHR is a suitable approach for the treatment of Graves’ disease.
